PATHOBIOLOGY IN FOCUS Experimental approaches to the study of cancer–stroma interactions: recent findings suggest a pivotal role for stroma in carcinogenesis

چکیده
An increasing body of research indicates that stroma surrounding cancer cells plays an important role in the development and subsequent behavior of the tumor. Studies using a wide range of techniques, including stromal cell isolation, modification of stromal-specific gene expression, and recreation of specific microenvironment conditions in culture, have demonstrated that stroma can promote cancer and that the expression patterns within the stroma can influence clinical outcome. Major hurdles in the study of the cancer stroma revolve around the cellular complexity of the tumor microenvironment, both in modeling the microenvironment and discovering/isolating pure populations of stromal cell types.

The role of the stroma in carcinogenesis.
This systematic review considers the most recent attitudes and news regarding the influence of the stroma on tumor initiation and progression. It is now widely accepted that tumor stroma plays an active role in carcinogenesis. Many different signaling molecules, ligands and signaling pathways recently have been discovered. This review considers the complexity of interactions between malignant c...

Research in recent years has accumulated a wealth of novel insight into mechanisms by which tumor cells interact with activated fibroblasts, endothelial cells, inflammatory and immune cells and the extracellular matrix. Cancer and stromal cells co-evolve throughout tumorigenesis. As a result, the tumor stroma is now regarded as an essential contributor to tumor establishment, progression and di...
متن کاملThe role of cancer-associated fibroblasts in breast cancer pathobiology.
The role of the tumor microenvironment (TME) is critical in cancer pathobiology. Of the components of the TME, cancer-associated fibroblasts (CAFs) play a major role. Breast cancer is a typical tumor type, forming abundant tumor stroma, and CAFs are involved in various aspects of breast cancer, including carcinogenesis, tumor progression, invasion, metastasis, inflammation, metabolism, therapy ...

Background and Aim: Prostate cancer is the second most prevalent malignancy in men, which affects health, economy, and social life. The ability to predict accurately, after prostatectomy, would increase optimization of disease management, because the presence or absence of basement membrane (BM) and reticular fibers are as important as cell lines in detecting of benignancy and malignancy of can...
متن کاملStromal Fibroblast in Age-Related Cancer: Role in Tumorigenesis and Potential as Novel Therapeutic Target
Incidence of most common cancers increases with age due to accumulation of damage to cells and tissues. Stroma, the structure close to the basement membrane, is gaining increased attention from clinicians and researchers due to its increasingly, yet incompletely understood role in the development of age-related cancer. With advanced age, stroma generates a pro-tumorigenic microenvironment, exem...
